Resumen

The present invention relates to an edible composition, such as an oral care composition, a medicament, or a food and feed composition, comprising a sorbitol oxidase enzyme and sorbitol.

Reivindicaciones

CLAIMS 1. An edible composition comprising a sorbitol oxidase enzyme and sorbitol. 2. A composition according to claim 1, wherein said composition further comprises xylitol. 3. A composition according to claim 1 or 2, wherein the sorbitol oxidase enzyme has a higher specific activity on sorbitol as compared to xylitol. 4. A composition according to claim 3, wherein the sorbitol oxidase enzyme has at least 1.5x higher specific activity on sorbitol as compared to xylitol 5. A composition according to claim 4, wherein the sorbitol oxidase enzyme has at least 2x higher specific activity on sorbitol as compared to xylitol 6. A composition according to any one of claims 1 to 5, wherein the sorbitol oxidase has a specific activity on sorbitol of at least 5 units/mg. 7. The composition according to any one of claims 1 to 6, wherein the specific activity of the enzyme on sorbitol and xylitol substrates is determined in vitro. 8. The composition according to any one of claims 1 to 6, wherein the specific activity of the sorbitol oxidase enzyme on sorbitol and xylitol substrates is determined in situ, within said composition. 9. The composition according to any one of claims 1 to 8, wherein the sorbitol present in the composition is between 0.05% wt% and 80wt%. 10. The composition according to any one of claims 2 to 9, wherein the xylitol present in the composition is between 0.05% wt% and 80wt%. 11. The composition according to any one of claims 2 to 10, wherein the combined sorbitol and xylitol present in the composition is between 0.05% and 80wt%. 12. The composition according to any one of claims 1 to 11, wherein the sorbitol oxidase enzyme is derived from a microbial strain of Streptomyces. 13. The composition according to any one of claims 1 to 12, wherein the sorbitol oxidase enzyme comprises a polypeptide sequence SEQ ID NO 1 or SEQ ID NO 2 or a homologue, variant or fragment thereof. 14. The composition according to any one of claims 1 to 13, wherein the sorbitol oxidase enzyme is present in said composition at an amount between 10 and 100,000 units per kilogram composition. 15. The composition according to any one of claims 1 to 14, wherein the oxidation of the sorbitol by the sorbitol oxidase produces or is capable of generating hydrogen peroxide in situ in the composition, and a first product. 16. The composition according to claim 15, wherein the first product is a sugar, such as glucose. 17. The composition according to claims 1 to 16, wherein said composition comprises a further enzyme which is capable of converting the first product to produce a further product. 18. The composition according to claim 17, wherein the further enzyme is a further oxidoreductase, which is capable of oxidizing the product to produce further hydrogen peroxide in situ in the composition and a further product, such as a sugar oxidase. 19. The composition according to claim 18, wherein the sugar-oxidase is selected form the group consisting of: carbohydrate oxidase, oligosaccharide oxidase, maltose oxidase, hexose oxidase, glucose oxidase, mannose oxidase, galactose oxidase, isolmaltulose oxidase, lactose oxidse, arabinose oxidase, erythrose oxidase, pentose oxidase, xylose oxidase, and triose oxidase, 20. The composition according to claim 18 or 19 wherein the sugar-oxidase is selected form the group consisting of: EC 1.1.3.4 glucose oxidase, EC 1.1.3.5 hexose oxidase, EC 1.1.3.9 galactose oxidase, EC 1.1.3.10 pyranose oxidase, EC 1.1.3.11 L- sorbose oxidase, and EC 1.1.3.40 D-mannitol oxidase. 21. The composition according to any one of the preceding claims wherein the amount of the at least one further oxidoreductase compared to the amount of sorbitol oxidase present in said composition is greater than 1, as measured by the respective number of enzyme units present in said composition. 22. The composition according to any one of the preceding claims, wherein the further product is or comprises a lactone. 23. The composition according to any one of claims 1 to 22, wherein the sorbitol oxidase is not obtained from Streptomyces IKD472, having a pH optimum at 7.5, is stable at pH 5.5 to 10.5 and at a temperature up to 65<0>C [as disclosed in JP 80892242]. 24. The composition according to any of claims 1 to 23, wherein the composition further comprises one or more additional enzymes selected from the group consisting of lactoperoxidase, laccase, peroxidase, lipase, acyl-transferase, mutanase, amylase, dextranase, protease, and lysozyme. 25. A oral care product comprising the composition according to any one of claims 1-25 and at least one further ingredient used in oral care products. 26. The oral care product according to claim 26, wherein the oral care product is in the form selected from the group consisting of a chewing gum, a mouthwash, a mouthspray, a pastille, a lozenge, a mouth freshener, a denture cleanser, a nasal spray, an oral paste, a sweetener, and a toothpaste. 27. An oral medicament product which comprises the composition according to any one of claims 1-25 or an oral care product according to claims 26 or 27 and at least one further ingredient used in oral medicament products. 28. A food or feed composition which comprises the composition according to any one of claims 1-25 and at least one suitable food or feed matrix material. 29. The food composition according to claim 28, wherein said food matrix is selected from the group consisiting of a dairy product, such as milk, cream, cheese, whey, yoghurt, butter; egg, such as egg yolk, egg white or whole egg; meat, such as processed meat, anaimal carcass and parts thereof, fish and fish products; beverages, such as fruit juice; fruits and vegetables; confectionary, such as candies, chocolates and mints. 30. The composition according to any of claims 1 to 24 for use as a medicament. 31. The use of a composition according to claims 1 to 24, in the manufacture of a medicament for the treatment or prevention of a medical disorder selected from gum disease, gingivitis, and periodontal disease. 32. The use of a composition according to claim any one of claims 1 to 24, or the oral care product according to claims 25 or 26 for a purpose selected from the group consisting of: Whitening and/or bleaching of teeth; inhibiting growth of oral bacteria; control of bad breath; prevention or treatment of gum disease such as gingivitis; prevention or reduction of dental plaque accumulation. 33. A method of medical treatment comprising administering the composition according to any one of claims 1 to 24, or the oral care composition according to claims 25 or 26, or the oral medicament according to claim 27 to a patient in need of treatment or prophylaxis. 34. The method according to claim 33, wherein the method is for the treatment or prevention of a medical disorder selected from gum disease, gingivitis, and periodontal disease. 35. A method for the preparation of a composition comprising admixing a sorbitol oxidase, and sorbitol andlxylitol and a suitable matrix, wherein said sorbitol oxidase is as defined in any one of claims 1 to 24. 36. The method according to claim 35, wherein said method further comprises the addition of at least one further enzyme such as a further oxidoreductase as defined in any one of claims 1 to 24. 37. A packaged product comprising the composition according to any one of claims 1 to 24, or the oral care product according to claim 25 or 26, or the medicament according to claim 27, or the food or feed product according to claim 28 or 29, wherein said composition is maintained in a oxygen limited environment within said packaged product so as to prevent or reduce the production of hydrogen peroxide within said packaged product. 38. A method for the generation of hydrogen peroxide in an edible composition, said method comprising the admixing of sorbitol oxidase and a polyol in an edible composition matrix, under suitable conditions and time to allow the generation of hydrogen peroxide and a first product. 39. The method according to claim 38, wherein a further enzyme, such as a further oxidoreducatse, is added to the edible composition matrix to reduce the accumulation or concentration of the first product, and optionally to produce further hydrogen peroxide. 40. A method for the preparation of hypothiocyanate in an edible composition, said method comprising methods according to claims 38 or 39, wherein the edible composition further comprises thiocyanate and lactoperoxidase, under suitable conditions and time to allow the generation of hypothiocyanatse from the thiocyanate and hydrogen peroxide produced by the methods according to claims 37 or 38. 41. The method according to any one of claims 38 - 40, wherein said method comprises exposing the composition contained within the packaged product according to claim 37 to an exogenous supply of oxygen.
