Resumen

The present invention relates to a method for determinating cytokine receptor, especially growth hormone receptor (GHR), activation, by the use of an antibody capable of discriminating between an activated and an inactive cytokine receptor conformation, especially GHR or growth hormone binding protein (GHBP) conformation, the method comprising: contacting the antibody and the cytokine receptor to form a complex, contacting a candidate ligand to thc complex, measuring the antibody binding to inactive cytokine receptor and thereby discriminating between an activated and an inactive conformation It also relates to a method for screening for a functionally cytokine agonists agonists and antagonists, especially functionally growth hormone (GH) agonists, among candidate ligands by the use of an antibody specific for activated cytokine receptor conformation. The invention also relates to an antibody directed to the hinge spanning subdomains I and II of the growth hormone receptor extracellular region and which is capable of discriminating between an activated and an inactive receptor conformation. The invention also relates to the use of the antibody for e.g. characterization of growth hormone receptor interactions and for screening for compounds stabilising the dimerization of GHR

Reivindicaciones

1. Method for determining human growth hormone receptor (hGHR) activation, by the use of a monoclonal antibody binding to hGHR or hGHBP and which binding is not inhibited by soluble hGH, and which is directed to the conformational epitope situated in the hinge spanning subdomains I and II of the hGHR extra cellular region and is capable of discriminating between an activated and an inactive hGHR, the method comprising the following steps: contacting the monoclonal antibody and hGHR to form a complex, contacting a candidate ligand to the complex, measuring the antibody binding to hGHR and thereby discriminating between an activated and an inactive conformation of hGHR. 2. Method according to claim 1 in which the hGHR extracellular region is human Growth Hormone Binding protein (hGHBP). 3. Method according to claim 1 or 2 for screening for a ligand capable of hGHR activation or inactivation. 4. Method according to claim 3 for screening for a human growth hormone (hGH) agonist or an hGH antagonist. 5. Method according to any of the claims 1-4 in which hGH is added.
